牙鲆MyoD基因单核苷酸多态性位点的筛选

IDENTIFICATION OF SINGLE NUCLEOTIDE POLYMORPHISMS OF MYOD IN JAPANESE FLOUNDER (PARALICHTHYS OVLIACEUS)

  • 摘要: 运用PCR-SSCP技术研究100尾牙鲆(Paralichthys olivaceus)MyoD基因的单核苷酸多态性(Single nucleotide polymorphisms, SNPs), 并将筛选到的突变位点与牙鲆生长性状进行相关性分析。结果表明, 在外显子1和内含子1上存在3个SNPs, 在外显子1 (MyoD2)基因座发现3种基因型AA、AB和BB, G863A突变, 属于同义突变。在内含子MyoD4基因座检测到DD、FF、CD、CE、DE和DF型个体。利用最小二乘法研究MyoD基因多态性位点对牙鲆生长性状的影响。结果表明, 外显子1的SNPs对生长性状无显著影响(P0.05)。内含子1的SNPs对牙鲆的生长性状影响均显著(P0.05)。研究结果为SNPs位点与牙鲆生长性能关联分析奠定了基础。

     

    Abstract: Single nucleotide polymorphisms (SNPs) of MyoD gene were analyzed in 100 Japanese flounder with PCR-SSCP Association between the polymorphisms and growth traits were analyzed. The results showed that the polymorphism sites were identified in exon 1(MyoD2) and intron 1(MyoD4). Three genotypes were found at the MyoD2 locus, denoted AA, AB and BB, and polymorphism site was G863A, being synonymous mutation. DD, FF, CD, CE, DE and DF genotypes were found at the MyoD4 locus. The association between different genotypes and Japanese flounder growth traits were analyzed by leastsquare analysis. The results indicated that there was no significant effect in the SNPs of exon 1 on growth traits (P0.05), the SNPs in intron 1 were significantly associated with the body weight, body length and body height (P0.05). These SNPs might be useful for a further research on MyoD genes relation to production performance in Japanese flounder.
