重金属Cu2+、Pb2+单因子及联合毒性对泥鳅卵细胞DNA的损伤效应

EFFECTS OF Cu2+ AND Pb2+ (SINGLE FACTOR AND JOINT TOXICITY) ON DNA DAMAGE IN MISGURNUS ANGUILLICAUDATUS OOCYTES

  • 摘要: 采用室内暴露试验方法, 研究了不同浓度Cu2+(0.01、0.10、0.25mg/L)、Pb2+(0.05、0.50、0.75mg/L)单因子染毒以及Cu2++Pb2+(0.01 mg/L+0.05mg/L、0.10 mg/L+0.50 mg/L、0.25 mg/L+0.75 mg/L)联合染毒对泥鳅卵细胞DNA的损伤效应, 并以SCGE技术进行检测。结果显示, Cu2+与Pb2+单因子染毒对泥鳅卵细胞DNA的损伤具有较为显著的剂量-效应与时间-效应关系(P0.05)。Cu2++Pb2+联合染毒, 在溶液暴露的0-5d表现为剂量-效应与时间-效应关系(P0.05); Cu2++Pb2+暴露5-10d 则表现出拮抗作用。研究结果显示, Cu2+、Pb2+ 单因子及联合染毒均造成泥鳅卵细胞DNA损伤, 具有基因毒性效应。

     

    Abstract: The effects of Cu2+ and Pb2+ (single factor and joint toxicity) attack on DNA damages in loach oosytes with different concentration of Cu2+(0.01, 0.10 and 0.25 mg/L respectively)and Pb2+(0.05, 0.50 and 0.75mg/L respectively), and Cu2++Pb2+(0.01 mg/L+0.05mg/L, 0.10 mg/L+0.50 mg/L and 0.25 mg/L+0.75 mg/L respectively)were studied by means of in door exposure and with the technique of SCGE. The results showed that the DNA damages in loach oosytes by single factor Cu2+ or Pb2+showed significant differences with dose-effect and time-effect (P0.05). As for joint attack of Cu2++Pb2, it showed that there were significant differences with dose-effect and time-effect under 5d exposure (P0.05), but it seemed to be an antagonism within 5d to 10d exposure. The results of the study showed that both single factor or joint attack by heavy metals will cause DNA damages to loach oosytes. Meanwhile, the genetic toxicity effects appears.
