卤虫脱壳卵的液氮冷冻保存研究

CRYOPRESERVATION OF DECAPSULATED ARTEMIA SP. EMBRYOS

  • 摘要: 作者以甘油、DMSO、葡萄糖和蔗糖为冷冻保护剂对卤虫脱壳卵进行了液氮冷冻保存研究。各保护剂对短期吸水发育的胚胎有一定的冷冻保护作用,在本实验条件范围内,高浓度保护剂较低浓度保护剂好,双重保护剂(甘油+DMSO、葡萄糖+蔗糖)冷冻保护作用较单一保护剂效果好。饱和NaCl溶液对各发育时期的胚胎均有较好的冷冻保护作用,但对后期胚胎的冷冻保护作用较弱。作者认为胚胎内冰晶的形成是卤虫脱壳卵冷冻致死的主要原因。冷冻保护剂对卤虫脱壳卵的冷冻保护作用可能与它们的脱水作用有关。

     

    Abstract: The decapsulated Anemia sp.embryos at their blastula stage were successfullycryopreserved in liquid nitrogen with various concentrations of glycerol,DMSO,glucose andsucrose as single cryoprotectants. Higher cryoprotectant concentrations were better than thelowers Dual cryoprotectants (glycerol+DMSO, glucose+sucrose)had stronger cryoprotectionthan single-type cryoprotection. Saturated NaCl solution showed a powerful cryoprotpetionfor various developmental stages of Anemia sp. embryos,although a weaker cryoprotCctionwas observed for later stage.Intraembryoic ice crystal formation was the main reason causing the death of the embryos.The cryoprotection of various cryoprotectants is possibly related to their dehydration effect.
