中华绒螯蟹各发育期APUD细胞的免疫细胞化学定位

IMMUNOCYTOCHEMICAL IDENTIFICATION AND LOCALIZATION OF APUD CELLS OF CHINESE MITTEN-HANDED CRAB ON DIFFERENT DIVELOPED STAGES

  • 摘要: 使用12种哺乳类、鱼类、昆虫激素为抗原制备的抗哺乳类血清,应用链霉亲和素-生物素过氧化物酶复合法免疫细胞化学技术对中华绒螯蟹1-5期状幼体、大眼幼体、幼蟹体内及成蟹消化系统的APUD细胞进行鉴别和定位.结果在1-5期状幼体及大眼幼体、幼蟹体内未见到典型的免疫活性阳性细胞;在成蟹消化系统中见到神经丝蛋白(NFP)、胰岛素(Ins)、血管活性肠肽(VIP)、降钙素(Cal)、五羟色胺(5-HT)、降钙素基因相关肽(CGRP)、生长激素(GH)、钙调素(CaM)及蜕皮激素(MH)等9种免疫活性在其除了后肠以外的不同部位有各自的阳性反应表现.而P物质(SP)、胰高血糖素(Glu)、神经特异烯醇酶(NSE)等3种抗血清在中华绒螯蟹成蟹消化系统中却未见其免疫活性阳性反应痕迹.本文着重描述以上9种免疫活性阳性反应在成蟹消化系统中分布情况及其阳性反应特征.

     

    Abstract: Using 12 kinds of antisera raised against mammalian from insects,fishes and mammalian,the APUD(amine pricursor uptake and decarboxylation)cells in the body of the first-fifth zoeal stages(Z1-Z5), a megalopal stage(M),a larva stage(L)of Chinese mitten-handed crab(Eriocheir sinensis)and in the digestive system of it's adult stage were identified and localized by using immunocytochemical technique of the strept avidin biotin-peroxidase complex method. The results showed that the immunoreactive positive cells in the body of the Z1-Z5,M and L of E. Sinensis were not found;9 kinds of antisera immunoreaction of nerve fiber protein,insulin,vasoactive intestinal polypeptide,calcitonin,5-hydroxytriptamine,calcitonin gene related peptide,growth hormone,calmodulin and molting hormone were found in the different parts of digestive system of E. Sinensis adult except hindgut. However 3 antisera of immunoreactive positive reaction of glucagon,substance P and neuron specific enuolase were not found in the digestive system of E. Sinensis adult. In present paper,the distribution of the above 9 kinds of immunoreactive positive reaction in the digestire system and their characteristics were described emphatically.
