PURIFICATION AND CHARACTERIZATION OF PHYCOERYTHRIN FROM RHODOSORUS MARINUS

  • 摘要: 从Rhodosorus marinus中提取了藻红蛋白,通过改进纯化方法,得到了三种电泳纯的藻红蛋白:B-型藻红蛋白1,B-型藻红蛋白2和b-型藻红蛋白(以下简称B-PE1,B-PE2和b-PE).分别测定这三种藻红蛋白聚合体和亚单位的分子量。测定了它们的可见光的吸收光谱和荧光光谱。两种B-PE的可见光的吸收光谱比b-PE的多一个498nm的吸收峰。三种藻红蛋白的氨基酸组成以酸性氨基酸和疏水性氨基酸为主,所以经等电聚焦测定的它们的等电点都是偏酸性的。

     

    Abstract: Three phycoerythrins ie B-phycoerythrinl(B-PE1),B-phycoerythrin 2(B-PE2)andb-phycoerythrin(b-PE)from Rhodosorus marinus were purified to electrophoresis purity:The molecular weights of the three phycoerythrins and their subunits were determined. Theabsorption spectra and the fluorescence spectra of the three phycoerythrins were measured.The absence or presence of an absorption peak at 498nm represents the major difference between the b-PE and B-PE.The amino acid compositions from the three phycoerythrins arerelatively rich in aliphatic and acidic amino acid. This is the reason why the isoelectric pointof the three phycoerythrins tends to acidity.
