末端标记限制性片段的基因组DNA指纹法

GENOMIC DNA FINGERPRINTING BY RESTRICTION FRAGMENT END LABELING

  • 摘要: 报道了分析基因组DNA多态性的方法:末端标记限制性片段的基因组DNA指纹法(RFEL-DNAFingerprinting)。通过此方法对莫桑比克非鲫核DNA进行了初步的分析。结果表明,此方法是一种简单、灵敏、稳定、可变换谱带复杂程度和有着广泛应用前景的一种鉴定生物基因型的方法。

     

    Abstract: A typical method, genomic fingerprinting by restriction fragment endlabeling (RFEL-DNA Fingerprinting) for fish was developed and applied toOreochromis mossambicus.Total genomic DNA was digested with a restrictionendonuclease.The fragments were end labeled with α-32p dATP by using theklenow fragment of DNA polymerase and separated by electrophoresis in 6% polyacrylamide/8M urea (sequencing gel).Depending on the different restrictionendonuclease, this method produced well-separated fragments (satellite DNA).Themethod described is a simple, sensitive, stcady and Powerful technique for genotypingfish species.
