    饲料异亮氨酸水平对豹纹鳃棘鲈生长、摄食及脂代谢的影响

    DIETARY ISOLEUCINE LEVELS ON GROWTH, FEEDING, AND LIPID METABOLISM OF LEOPARD CORAL GROUPER (PLECTROPOMUS LEOPARDUS)

    • 摘要: 本研究旨在探究饲料异亮氨酸(Isoleucine, Ile)水平对豹纹鳃棘鲈(Plectropomus leopardus)生长、摄食及脂代谢的影响。实验设计了1.96%、2.11%、2.29%、2.40%、2.53%、2.68%和2.84% (占饲料干重)七种异亮氨酸水平饲料, 分别缩写为Ile1.96、Ile2.11、Ile2.29、Ile2.40、Ile2.53、Ile2.68和Ile2.84, 每个异亮氨酸水平组含3个重复。实验鱼初始体重为(34.93 ± 0.27) g, 15尾/重复, 养殖9周。结果显示, Ile2.84组增重率(WGR)、每日摄食量(DFI)、特定生长率(SGR)和蛋白质效率(PER)显著低于其他组(P<0.05), 而饲料系数(FCR)显著高于其他组(P<0.05)。以WGR为评价指标, 经双斜率折线模型分析表明, 当饲料异亮氨酸水平超过2.68%时, 豹纹鳃棘鲈的生长出现下降。Ile2.84组肝体比(HSI)、肥满度(CF)及全鱼、肌肉、肝脏脂肪含量显著低于其他组(P<0.05), 血浆甘油三酯(TG)、总胆固醇(T-CHO)、低密度脂蛋白胆固醇(LDL-C)含量表现出类似变化趋势(P<0.05), 而高密度脂蛋白胆固醇(HDL-C)含量表现出相反变化趋势(P<0.05)。Ile2.84组脑组织促食欲基因npy及肠道组织抑食欲相关基因(t1r1t1r3、cck)的表达水平显著高于其他组(P<0.05); 与其他组相比, Ile2.84组肝脏中脂肪分解相关基因(ampkαpparαcpt-1hsl)的表达水平显著增加(P<0.05), 但脂肪合成相关基因(acc1α和fas)的表达水平显著降低(P<0.05)。总之, 配合饲料中1.96%—2.68%的异亮氨酸水平(占饲料蛋白的3.80%—5.17%)能够满足豹纹鳃棘鲈生长的需求。

       

      Abstract: This study investigated the effects of dietary isoleucine (Ile) levels on growth, feeding, and lipid metabolism of leopard coral grouper (Plectropomus leopardus). Seven experimental diets, each with three replicates, were formulated with graded Ile levels of 1.96%, 2.11%, 2.29%, 2.40%, 2.53%, 2.68%, and 2.84% of dry matter, designated as Ile1.96, Ile2.11, Ile2.29, Ile2.40, Ile2.53, Ile2.68, and Ile2.84, respectively. Juvenile fish (initial body weight of 34.93±0.27 g) were stocked at 15 fish per replicate, with three replicates per diet, and fed for 9 weeks. Results showed that the Ile2.84 group exhibited significantly lower weight gain rate (WGR), daily feed intake (DFI), specific growth rate (SGR), and protein efficiency ratio (PER) compared with all other groups (P<0.05), while its feed conversion rate (FCR) was significantly higher (P<0.05). The analysis of two-slope broken-line model on WGR indicated that growth declined when dietary isoleucine levels exceeded 2.68%. The Ile2.84 group exhibited significantly lower hepatosomatic index (HSI), condition factor (CF), and crude lipid content in whole body, muscle, and liver compared to other groups (P<0.05). Consistently, plasma levels of triglycerides (TG), total cholesterol (T-CHO), and low-density lipoprotein cholesterol (LDL-C) were significantly decreased (P<0.05), while high-density lipoprotein cholesterol (HDL-C) levels exhibited the opposite trend (P<0.05). Ile2.84 group showed significantly higher expression of brain orexigenic genes (npy) and intestinal anorexigenic genes (t1r1, t1r3, and cck) compared to other groups (P<0.05). The Ile2.84 group exhibited significantly reduced expression levels of hepatic lipid synthesis-related genes (acc1α and fas), but significantly increased expression levels of lipid catabolism-related genes (ampkα, pparα, cpt-1, and hsl) (P<0.05). In conclusion, dietary isoleucine level ranging from 1.96% to 2.68% (3.80%–5.17% of dietary protein) are sufficient to support optimal growth in P. leopardus.
