    受控条件下人工模拟产卵环境诱导中华鲟自然繁殖后代的亲权分析

    PARENTAL ANALYSIS OF CHINESE STURGEON OFFSPRING DERIVED FROM NATURAL REPRODUCTION INDUCED IN A SIMULATED SPAWNING ENVIRONMENT UNDER CONTROLLED CONDITIONS

    • 摘要: 为评估2025年模拟生境诱导下中华鲟人工培育群体的产卵行为、交配策略及子代遗传贡献, 本研究采用线粒体D-loop测序与19个微卫星位点对25尾亲本75尾子代进行亲权鉴定, 分析亲本参与度及家系结构。结果表明: 25 尾亲鱼中共有2尾雌鱼(25%)和4尾雄鱼(23.5%)成功参与繁殖并产生4个家系, 其中一批次为“一雌一雄”单对配对(1162♀×1549♂), 另一批次表现为“一雌多雄”的混交模式。其中核心母本(1197♀)的后代贡献率达81.9%, 分子标记对子代的成功分配率为96%。参与繁殖的6尾亲本19个微卫星位点的平均等位基因数Na、平均观测杂合度Ho、平均期望杂合度He及平均多态信息含量PIC分别为7.37、0.740、0.800和0.769, 整体遗传多样性较好, 75尾子代个体19个微卫星位点遗传多样性数据与6尾亲本接近, 后代群体基本保持了亲本群体的遗传多样性。本研究证实了中华鲟在受控条件下自然繁殖过程中采取了一雌一雄或一雌多雄的混合交配策略, 为优化中华鲟栖息地修复技术及实现野外种群的自我维持提供了重要的技术支撑与理论指导。

       

      Abstract: In order to evaluate the spawning behavior, mating strategies, and genetic contribution of artificially reared Chinese sturgeon population under simulated habitat conditions in 2025, this study employed mitochondrial D-loop sequencing combined with 19 microsatellite loci to perform parentage identification on 25 parental fish and their 75 offspring, and analyzed the parental involvement and pedigree structure. Results indicated that among the 25 parental fish, 2 females (25%) and 4 males (23.5%) successfully participated in reproduction, resulting in the formation of 4 pedigrees; one batch exhibited a “one female–one male” single-pair mating pattern (1162♀ × 1549♂), while another batch followed a “one female–multiple males” hybrid mating pattern. The offspring contribution rate from the core maternal line (1197♀) reached 81.9%, and the success rate of molecular markers in assigning offspring to their parents was 96%. For the 6 parental fish involved in reproduction across the 19 microsatellite loci, the average number of alleles (Na), average observed heterozygosity (Ho), average expected heterozygosity (He), and average polymorphism information content (PIC) were 7.37, 0.740, 0.800, and 0.769, respectively, indicating relatively high overall genetic diversity. Genetic diversity data obtained from the 75 offspring individuals were consistent with those from the 6 parental fish, demonstrating that the offspring population retained the genetic diversity of the parental population. This study confirms that Chinese sturgeons adopt either a “one female–one male” or a “one female–multiple males” hybrid mating strategy during natural reproduction under controlled conditions, providing crucial technical support and theoretical guidance for optimizing habitat restoration techniques for Chinese sturgeons and achieving the self-sustenance of wild populations.
