    吲哚-3-丙酸对大黄鱼稚鱼生长性能和肠道健康的影响

    DIETARY INDOLE-3-PROPIONIC ACID ON THE GROWTH PERFORMANCE AND INTESTINAL HEALTH OF LARGE YELLOW CROAKER (LARIMICHTHYS CROCEA) LARVAE

    • 摘要: 为探究饲料中添加吲哚-3-丙酸(IPA)对大黄鱼稚鱼生长性能和肠道健康的影响, 以初始均重(4.98±0.07) mg的大黄鱼稚鱼为研究对象, 投喂饲料中IPA添加量分别为0、0.005%、0.010%和0.020%的微颗粒饲料, 进行为期30d的摄食生长实验。结果表明, 在生长性能方面, 与对照组相比, 饲料中添加0.005%和0.010% IPA均显著提高了稚鱼的存活率、特定生长率、终末体长和终末体重。在肠道发育方面, 与对照组相比, 饲料中添加0.005%和0.010% IPA均显著提高了肠道褶皱高度。同时, 饲料中添加0.005% IPA显著提高了肠道刷状缘麦芽糖酶活力; 添加0.010%和0.020% IPA显著提高了刷状缘亮氨酸氨基肽酶活力, 促进了肠道发育。在肠道发育相关基因方面, 与对照组相比, 饲料中添加0.005% IPA显著上调了pcnaodccateninα1zo-1zo-2claudin1claudin5occludin的表达水平。在消化能力方面, 与对照组相比, 饲料中添加0.005% IPA显著提高了肠道淀粉酶、脂肪酶和胰蛋白酶活力。在吸收能力方面, 与对照组相比, 饲料中添加0.005% IPA显著提高了肠道刷状缘Na+/K+-ATP酶活力。在消化吸收相关基因方面, 与对照组相比, 饲料中添加0.005% IPA显著上调了pept1slc7a7slc7a5slc6a6slc7a1aslc36a1的mRNA表达水平; 饲料中添加0.010% IPA显著上调了cckpept1slc6a6slc36a1的mRNA表达水平, 提高了稚鱼的消化吸收能力。研究证实了饲料中添加适量的IPA能够显著提高大黄鱼稚鱼的生长性能, 综合各项指标, IPA的适宜添加量为0.005%—0.010%。

       

      Abstract: To investigate the effects of dietary isophorone (IPA) on the growth performance and intestinal health of large yellow croaker (Larimichthys crocea) larvae, a 30d feeding trial was conducted. Larvae with an initial mean weight of (4.98±0.07) mg were selected as research subjects and fed micro-diets supplemented with IPA at levels of 0, 0.005%, 0.010%, and 0.020%. Regarding survival and growth performance, supplementation with 0.005% and 0.010% IPA significantly enhanced the survival rate, specific growth rate, final body length, and final body weight compared with the control group. In terms of intestinal development, a significant increase in intestinal fold height was observed in the 0.005% and 0.010% IPA groups relative to the control. Concurrently, dietary supplementation of 0.005% IPA significantly increased intestinalbrush border lactase activity; meanwhile, addition of 0.010% and 0.020% IPA significantly enhanced brush border leucine aminopeptidase activity, whereby promoting intestinal development. In terms of genes related to intestinal development, the mRNA expression levels of pcna, odc, cateninα1, zo-1, zo-2, claudin1, claudin5, and occludin were significantly upregulated by 0.005% IPA compared with the control. In terms of digestive capacity, the activities of intestinal amylase, lipase, and trypsin were significantly improved by 0.005% IPA. Regarding absorptive capacity as reflected by brush border enzyme activities, the activity of intestinal Na+/K+-ATPase was significantly increased by the 0.005% IPA treatment. In terms of digesting and absorbing related genes, the mRNA expression levels of pept1, slc7a7, slc7a5, slc6a6, slc7a1a, and slc36a1 were significantly upregulated by the 0.005% IPA level; meanwhile, the mRNA expression levels of cck, pept1, slc6a6, and slc36a1 were significantly upregulated by the 0.010% IPA level, thereby enhancing the digestive and absorptive capacities of the larvae. That the growth performance of L. crocea larvae could be significantly enhanced by dietary IPA supplementation was demonstrated in this study. Consistent with the comprehensive analysis of all parameters, the optimal addition level of IPA was suggested to be 0.005%—0.010%.
