福建沿海常见鲸类DNA条形码库的建立及应用分析

ESTABLISHMENT AND APPLICATION ANALYSIS OF A DNA BARCODE REFERENCE DATABASE FOR COMMON CETACEANS ALONG THE FUJIAN COAST

  • 摘要: 为了更有效地进行鲸类物种识别, 本研究以2002—2020年间福建沿海搁浅的51头鲸豚为研究对象, 联合线粒体COⅠ基因与D-loop区, 初步建立了该区域的常见鲸类DNA条形码数据库。最终结合形态学鉴定与分子技术, 成功鉴定出6科10属14种鲸豚, 其中包括罕见的深潜物种-银杏齿中喙鲸(Mesoplodon ginkgodens)。此外, 通过遗传距离和系统发育树等方法分析了COⅠ 基因和D-loop区作为条形码在福建沿海常见鲸豚类物种鉴定中的适用性。本数据库的初步建立不仅进一步完善了福建海域鲸类物种的信息资料, 为鲸类物种形态鉴定难题提供了解决方案, 还将为鲸类等海洋哺乳动物的保护管理及种群监测奠定研究基础和提供技术支持。

     

    Abstract: The western coast of Taiwan Strait in Fujian has abundant marine biomass and serves as a hotspot for cetacean diversity. Accurate taxonomic delineation of these endangered mammals is critical for their conservation, which is commonly dependent on carcass decomposition degree and observer expertise. To circumvent these limits, we constructed a regional DNA barcode reference database including mitochondrial COI and D-loop as markers for fifty-one stranded cetaceans collected along the Fujian coast between 2002 and 2020. Based on the morphological inspection and genetic verification, we successfully identified fourteen species belong to ten genera and six families, including a rarely reported Ziphiid species (Mesoplodon ginkgodens). Phylogenetic reconstruction and genetic distance analysis confirmed that application the COI and D-loop as species identification barcode for common cetaceans along the Fujian coast. The established barcode database not only refines the documented cetacean diversity in this region but also serves as a valuable supplementary tool for traditional morphological identification. This work further offers a primary foundation for long-term population monitoring and conservation management of marine mammals in the Taiwan Strait.
