持续高温胁迫对克氏原螯虾生长、消化酶活性与免疫指标的影响

EFFECTS OF SUSTAINED HIGH-TEMPERATURE STRESS ON GROWTH PERFORMANCE, DIGESTIVE ENZYME ACTIVITIES AND IMMUNE INDEXES OF CRAYFISH, PROCAMBARUS CLARKII

  • 摘要: 为探讨持续高温胁迫对克氏原螯虾(Procambarus clarkii)生长及健康的影响, 分别在适宜温度(24℃, 对照)和不同高温胁迫温度(29℃、32℃和35℃)条件下对该虾体重(7.19±0.29) g进行了为期30d的养殖试验。结果显示: (1)随着胁迫温度的升高克氏原螯虾的成活率(SR)显著下降(P<0.05); 终末体重(FBW)、终末体长(FBL)、增重率(WGR)及特定生长率(SGR)等生长指标随着温度呈先升高后降低的趋势。在29℃时生长性能指标达到峰值并显著高于其他3个温度组(P<0.05); 与24℃相比, 32℃的高温胁迫对该虾的生长性能未造成显著影响(P>0.05)而35℃的高温则使其生长性能显著下降(P<0.05); (2)胃蛋白酶、淀粉酶和脂肪酶活性随着温度升高呈先升高后降低的趋势。在29℃时3种消化酶活性达到峰值并显著高于其他3个温度组(P<0.05), 在32℃时淀粉酶和脂肪酶的活性显著高于对照组(P<0.05), 在35℃时胃蛋白酶活性显著低于对照组(P<0.05); (3)在高温胁迫条件下, 随着温度上升血淋巴中的酸性磷酸酶(ACP)和碱性磷酸酶(AKP)的活性显著降低, 而谷草转氨酶(AST)和谷丙转氨酶(ALT)的活性逐步显著升高(P<0.05); 肝胰腺的总抗氧化能力(T-AOC)和总超氧化物歧化酶(T-SOD)活性在29℃时最低(P<0.05)且T-SOD活性显著低于其他温度组(P<0.05), 而丙二醛(MDA)含量随温度上升而逐步显著升高(P<0.05); 肝胰腺和鳃组织的热应激蛋白基因(HSP70)相对表达量在29℃时显著上升(P<0.05)且在35℃时最高(P<0.01)。结果表明, 29℃的持续高温虽可显著促进克氏原螯虾的生长与消化但对其免疫指标显著不利, 而32℃及以上的持续高温胁迫则严重影响其存活与生长、消化、免疫及抗氧化功能。

     

    Abstract: In order to investigate the effects of sustained high-temperature stress on the growth performance and health status of crayfish, Procambarus clarkii, the P. clarkii of (7.19±0.29) g were cultured at the suitable temperature (24℃, the control) and the higher temperatures (29℃, 32℃ and 35℃) for 30 days, respectively. The results showed as follows: (1) With the increase of the stress temperature, the survival rate (SR) of P. clarkii decreased significantly (P<0.05); the growth performance indexes including final body weight (FBW), final body length (FBL), weight gain rate (WGR) and specific growth rate (SGR) increased firstly and then decreased with the rise of temperature. The growth performance indexes reached the peak at 29℃ and was significantly higher than those of other treatments (P<0.05); compared with 24℃, 32℃ of high-temperature stress had no significant effect on the growth performance (P>0.05), but the growth performance decreased significantly under 35℃ of high-temperature stress (P<0.05); (2) The activities of pepsin, amylase and lipase firstly increased and then decreased with the increasing temperature. The activities of three digestive enzymes reached the peak at 29℃ and were significantly higher than those of other treatments (P<0.05), the activities of amylase and lipase at 32℃ were significantly higher than those of the control (P<0.05), and the pepsin activity at 35℃ was significantly lower than that of the control (P<0.05); (3) Under the conditions of high-temperature stress, the activities of acid phosphatase (ACP) and alkaline phosphatase (AKP) in serum significantly decreased while the activities of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) gradually increased significantly with the increasing temperature (P<0.05); The total antioxidant capacity (T-AOC) and total superoxide dismutase (T-SOD) activity in hepatopancreas were the lowest at 29℃ (P<0.05), and the T-SOD activity was significantly lower than those of other treatments (P<0.05), while the malondialdehyde (MDA) contents gradually increased with increasing temperature (P<0.05). The relative expression of heat stress protein gene (HSP70) of hepatopancreas and gill increased significantly at 29℃ (P<0.05) and was the highest at 35℃ (P<0.01). The results revealed that the 29℃ of sustained high-temperature significantly promote the growth performance and the digestion but significantly affects the immune function of P. clarkii adversely, and the sustained high-temperature stress of 32℃ and above has serious detriment to its survival and growth, digestion, immune and antioxidant functions.
