利用胶束电动毛细管电泳比较五种提取鲫鱼总蛋白方法研究
COMPARISON OF FIVE EXTRACTION METHODS BY ANALYZING THE TOTAL PROTEINS FROM CARP USING EMKC

摘要: 毛细管电泳又称高效毛细管电泳(High2performance capillary electrophoresis,HPCE),是20 世纪80 年代问世的一种高效液相分离法。毛细管电泳分离效率高,样品用量少,灵敏度高,快速,分析范围广,广泛应用于离子、小分子、糖类、核酸、多肽、蛋白、病毒、细菌的分离。本验采用的是胶束电动毛细管电泳(Micellar electrokinetic chromatography,EMKC),基本原理是利用溶质在胶束与水相间分配系数差异达到分离目的,不但能够分离带电物质,而且能够分析不带电荷的中性物质。因此EMKC 常被用于蛋白质和多肽的分离,如Nishi H1等利用EMKC 电泳成功分离了β-内酰胺抗菌素(β2lactam antibiotics) 。
