鳜及大眼鳜线粒体DNA比较研究

COMPARATIVE STUDIES ON MITOCHONDRIAL DNA FROM SINIPERCA CHUATSI AND SINIPERCA KNERI

  • 摘要: 采用10种限制性内切酶HindⅢ、EcoRⅠ、BglⅠ、BglⅡ、BamHⅠ、PvuⅡ、PstⅠ、SalⅠ、XbaⅠ、XhoⅠ对鳜及大眼鳜肝脏线粒体DNA(mtDNA)进行了分析。鳜鱼mtDNA分子量为9.703×106u,大小为16.19kb;大眼鳜mtDNA分子量为9.603×106u,大小为16.02kb。根据单、双酶切结果构建了鳜及大眼鳜mtDNA 10种酶限制性酶切图谱,并对两种鱼的mtDNA酶切图谱进行了比较。

     

    Abstract: Using the method of density gradient centrifugation and DNase I, RNase digestion, We have isolated and purified the mtDNA from Siniperca chuatsi and.Siniperca kneri.The mtDNA from the fishes have been digested with the following 10 kinds of restriction endonucleases, EcoRⅠ, Hind Ⅲ, Bgl Ⅱ, BamHⅠ, Bgl Ⅰ,Sal Ⅰ, Pst Ⅰ, Pvu Ⅱ, Xba Ⅰ and Xho Ⅰ.The molecular weights of Siniperca chuatsi and Siniperca kneri are 9.703×106u, 16.19kb and 9.603×106u, 16.02kb,respectively.We have constructed the restriction maps of 10 kinds of endonucleases of the mtDNA from Siniperca chuatsi and Siniperca kneri.
