我国产日本医蛭水蛭素的分离和纯化

ISOLATION AND PURIFICATION OF HIRUDIN FROM A BLOOD-SUCKING LEECH,HIRUDO NIPPOIA,IN CHINA

  • 摘要: 采用离子交换柱层析与凝胶过滤柱层析的方法对从我国产的日本医蛙Hirudo nipponica(whitman)活体中提取出的唾液腺分泌物进行分离和纯化,获得了较纯净的水蛭素(Hirudin)。测定结果表明:日本医蛭唾液腺分泌物提取液中水蛭素含量为4AT-U/ml;纯化总得率为15%;纯化后的产物比活力为6708AT-U/mg蛋白。

     

    Abstract: This paper deals with the isolation and purification of hirudin from a bloodsucking leech, Hirudo nipponia in China. Essentially, the experimental process requires an anion exchange chromatography on DEAE-C52 and a gel filtration chromatography on Sephadex-G25. The hirudin activity were measured by the method of titration with thrombin. The salivary gland secretions have been obtained from the alive leeches. The hirudin content in salivary gland secretions is 4 AT-U/ml;the hirudin production rate is 15%; ratio activity after purifing is 6708 AT-U/ml protein.
