POLYMORPHISM AND RESTRICTION MAP OF MITOCHONDRIAL DNA FROM CLARIAS FUSCUS

  • The mtDNA from liver of Clarias fuscus has been analysed with 8 kinds of restriction endonucleases. Xho Ⅰ. EcoRⅠ. PstⅠ. BamHⅠ. Xba Ⅰ. HindⅢ have 2. 3. 1. 1. 3 and 5 sites on this mtDNA, BglⅠ and BglⅡ have 2 sites on mtDNA (type Ⅰ), and 1 site on mtDNA (type Ⅱ). There was length polymorphism in liver mtDNA of C. fuscus. Two restriction types were observed, when digested with Bgl Ⅰand BglⅡrespectively. The molecular weight of C. fuscus mtDNA is 10.242×106u, and length is about 16.68 kilobase pairs. The author established the restriction maps of C. fuscus mtDNA by double digestion, and analysed the restriction fragment length polymorphisms (RFLP) of liver mtDNA in C. fuscus.
